Composition
Sea salts (SIGMA) 35 g   Wolfe's mineral elixier* 1 ml   NH4Cl 0.1 g   KH2PO4 0.05 g   Yeast extract 0.5 g   Trypticase Peptone (BBL) 0.25 g   Sodium glutamate 0.1 g   Na2CO3 0.5 g   Vitamin solution** 10 ml   Distilled water 1 L   pH 7.4-7.8       Dissolve ingredients except for carbonate and vitamins and sparge solution with N2/CO2 (80/20) gas mixture until it is anoxic and saturated with CO2 (at least 30 min.). Dispense solution in vials suitable for anaerobic cultures to 50% of volume under air atmosphere. Close vials with butyl rubber septa to prevent free exchange of oxygen with the external atmosphere and autoclave. Add vitamins from a sterile anoxic stock solution prepared under N2 and carbonate form an autoclaved stock solution prepared under N2/CO2 gas mixture. The vitamin solution should be sterilized by filtration.
