Composition
NH4Cl 1 g   MgCl2·6H2O 0.1 g   CaCl2·2H2O 0.1 g   K2HPO4 0.4 g   Bacto Yeast Extract (Difco) 2 g   Polypeptone 2 g   Resazurin 1 mg   Carbonate mixture* 210 ml   Tris-HCl buffer** 10 ml   Vitamin solution*** 10 ml   Trace elements solution**** 10 ml   Cysteine-HCl 0.3 g   Na2S·9H2O 0.3 g   Distilled water 760 ml     Mix ingredients except for carbonate mixture, Tris-HCl buffer, vitamin solution, cysteine-HCl and Na2S·9H2O, and autoclave under a H2 atmosphere. Carbonate mixture, Tris-HCl buffer, cysteine-HCl (3% solution) and Na2S·9H2O (3% solution) are separately autoclaved under a N2 atmosphere. Prior to inoculation, add filter-sterile vitamin solution, carbon mixture, Tris-HCl buffer, cysteine-HCl and Na2S·9H2O. Pressurize the inoculated vessels to 150 kPa with H2.                   *Carbonate mixture       NaHCO3 10 g       Na2CO3 0.5 g       Distilled water 210 ml     Mix ingredients and autoclave under a N2 atmosphere.                   **Tris-HCl buffer     Dissolve 2 M Tris, adjust pH to 8.4 with HCl, and autoclave under a N2 atmosphere.                   ***Vitamin solution       Biotin 2 mg       Folic acid 2 mg       Pyridoxine-HCl 10 mg       Thiamine-HCl 5 mg       Riboflavin 5 mg       Nicotinic acid 5 mg       Ca-pantothenate 5 mg       p-Aminobenzoic acid 1 mg       Vitamin B12 0.01 mg       Distilled water 1 L                   ****Trace elements solution       2Na-EDTA 0.64 g       MgSO4·7H2O 6.2 g       MnSO4·4H2O 0.55 g       NaCl 1 g       FeSO4·7H2O 0.1 g       CoCl2·6H2O 0.17 g       CaCl2·2H2O 0.13 g       ZnSO4·7H2O 0.18 g       CuSO4 0.05 g       KAl(SO4)2·12H2O 0.018 g       H3BO3 0.01 g       Na2MoO4·2H2O 0.011 g       NiCl2·6H2O 0.025 g       Na2SeO4 0.004 g       Na2WO4 0.004 g       Distilled water 1 L     Dissolve the 2Na-EDTA in 500 ml of distilled water, and after adding the salts, make up to 1 L.
