Composition
Solution A* 500 ml   Solution B** 500 ml       Agar solid medium of GYS contained 2% agar. In order to prevent the hydrolysis of agar and glucose by autoclaving at low pH condition, Solutions A and B are autoclaved separately and combined before solidified (50-60℃).                   *Solution A       KH2PO4 1 g       (NH4)2SO4 1 g       Yeast extract 0.3 g       MgCl2·6H2O 0.2 g       CaCl2 45 mg       NaCl 0.2 g       Trace element*** 1 ml       Distilled water 500 ml       Adjust pH to 3.5-5.0 with 1 N H2SO4 for each strain (see Comments in catalogue) .                   **Solution B       Glucose 2 g       Agar (if needed) 15 g       Distilled water 500 ml                   ***Trace element solution       2Na-EDTA 5 g       FeCl2·4H2O 1 g       H3BO3 0.3 g       CoCl2·6H2O 0.11 g       ZnCl2 50 mg       MnCl2·4H2O 30 mg       Na2MoO4·2H2O 30 mg       NiSO4·6H2O 30 mg       CuCl2·2H2O 10 mg       Distilled water 1 L
