| | Mix ingredients except vitamin B12, bicarbonate and sulfide, and dispense the medium into suitable culture vessels under a stream of nitrogen gas, seal with butyl rubber stoppers. After autoclaving (121°C, 15 min), aseptically and anaerobically add vitamin B12 solution, bicarbonate solution and sulfide solution using sterile syringes. The bicarbonate and vitamin solutions should be sterilized by filtration. Adjust the pH of the medium to pH 7. |
