Modified synthetic Crenarchaeota medium for strain SCM1
Composition
  NaCl 26 g
  MgSO4·7H2O 5 g
  MgCl2·6H2O 5 g
  CaCl2·2H2O 1.5 g
  KBr 0.1 g
  HEPES buffer* 10 ml
  1 M NaHCO3 buffer** 5 ml
  KH2PO4 solution*** 5 ml
  FeNaEDTA solution**** 5 ml
  Trace element solution***** 1 ml
  NH4Cl solution****** 2.5 ml
  Sodium alpha-Ketoglutamate solution******* 5 ml
  Distilled water 1 L
 
    Mix ingredients except for HEPES buffer, NaHCO3 buffer, KH2PO4 solution, FeNaEDTA solution, trace element solution, NH4Cl solution and alpha-ketoglutamate solution, and autoclave at 121°C for 20 min. HEPES buffer, and KH2PO4, FeNaEDTA, trace element, and NH4Cl solutions are sterilized separately by autoclaving and added just before inoculation. NaHCO3 buffer and alpha-ketoglutamate sollution are also added just before inoculation after filter-sterile. Final pH should be 7.4-7.6.
             
    *HEPES buffer
      NaOH 24 g
      HEPES 238.4 g
      Distilled water 1 L
 
    Dissolve NaOH, and then add HEPES slowly. Adjust pH to 7.6 with NaOH solution. Sterilize by autoclaving.
             
    **1 M NaHCO3 buffer
 
    Dissolve NaHCO3, and sterilize by filter.
             
    ***KH2PO4 solution
 
    Dissolve KH2PO4 (0.4 g/L=2.93 mM), and sterilize by autoclaving.
             
    ****FeNaEDTA solution
 
    Dissolve FeNaEDTA (578 mg/L=1.5 mM), and sterilized by autoclaving. Store at 4°C.
             
    *****Trace element solution
      HCl (35%) 9 ml
      MnCl2·4H2O 0.1 g
      CoCl2·6H2O 0.19 g
      ZnSO4·7H2O 0.144 g
      CuCl2·2H2O 0.002 g
      H3BO3 0.03 g
      Na2MoO4·2H2O 0.036 g
      NiCl2·6H2O 0.024 g
      Distilled water 1 L
 
    Add HCl, dissolve trace elements, and sterilize by autoclaving. Store at 4°C.
             
    ******NH4Cl solution
 
    Dissolve NH4Cl (0.4 M), and sterilize by autoclaving.
             
    *******Sodium alpha-ketoglutamate solution
 
    Dissolve alpha-ketoglutamate (20 mM), and sterilize by filter. Store at 4°C.
