Composition
Bacto Yeast Extract (Difco) 1 g   NaCl 30 g   K2HPO4 90 mg   KH2PO4 70 mg   CaCl2·2H2O 1.06 g   MgCl2·6H2O 4.18 g   KCl 0.33 g   NH4Cl 0.535 g   NaHCO3 2 g   Na2S·9H2O 0.3 g   Cysteine-HCl 0.3 g   Trace element solution* 10 ml   Vitamin solution** 10 ml   Resazurin 1 mg   Distilled water 1 L   pH 7       For vial cultures, dissolve all ingredients except for yeast extract, vitamin solution, Na2S·9H2O and cysteine-HCl (up to 900 ml), flush N2/CO2 (80/20) (pH 7), dispense 19 ml into 50 ml vials, seal with butyl rubber stoppers with flushing N2/CO2 (80/20) and autoclave. Add 0.5 ml of each of filter-sterilized Na2S·9H2O solution (0.3 g Na2S·9H2O/50 ml) and cysteine-HCl solution (0.3 g cysteine-HCl/50 ml) per each vial separately. Add 0.5 ml of filter-sterilized 40 g/l yeast extract solution. Inoculum size 5%.
